- How tumor cells maintain proliferative status in low ecdysone/insulin environments. By employing a p53 induced senescence tumorigenesis model, we are studying a molecular network that increases tumor cell susceptibility to proliferation under limited hormonal conditions. We demonstrate that senescent-induced tumors become extremely sensitive to the low ecdysone/insulin levels present in their environment through the acquisition of a juvenile state mediated by the transcription factor Chinmo and activation of the mTOR pathway. We will study in detail how the transcription factor Chinmo regulates its target genes to promote insulin and ecdysone resistance in the tumors.
