- The CD40 receptor and its ligand (CD40L) is one of the key molecular pairs of the stimulatory immune-checkpoints, playing a significant role in inflammatory and autoimmune diseases. Related to the adaptive immune responses, CD40 stimulation boosts the B cell activation initiated by antigen recognition, leading to an effective humoral immune response against pathogens, and its deficiency causes the absence of germinal centres and low levels of circulating immunoglobulins. We have recently identified Diacylglycerol-kinase zeta (DGKz) as an essential mediator of CD40 functions in the B cell immune response. We found that DGKz supports mTORC1 activation, mitochondrial remodelling, cell proliferation and the transcriptional network that determines B cell fate downstream of CD40 stimulation. The lack of DGKz leads to decreases in germinal centre responses and generation of long-lived plasma cells and memory B cells in vivo. This project pursues to further explore the CD40/DGKz axis in homeostasis and pathology, in particular in metabolism, epigenetics and transcription factors related aspects.
